isolation and identification of polyethylene degrading bacteria from sludge

Plastic wastes accumulating in the environment are posing an ever increasing ecological threat. Plastics are used in our daily life they are unable to degrade for years remain in environment as same. Now a days biodegradable plastics are used in daily life because they are easily degradable. They are considered as an eco-friendly. They have an increasing range of potential application and are driven by the growing use of plastics in packaging.int this study plastic degrading microorganisms isolated and identified from Peshawar sewage area. The samples were inoculated on nutrient agar media (general purpose media), blood agar media (for haemolysis), Eosin Methylene Blue Agar media (for E.coli, sheen green colonies) and MacConkey agar media (for Enterobacteriaceae gram negative LF and NLF) for isolation of bacterial species. For the identification of bacterial species oxidase test, catalase test, TSI test, indole test urease and citrate tests were performed. For confirmation of gram negative species API kits were used. For making the plastics hydrophobic pre-treatment was performed ( which include soak plastic in distilled water for 20 minutes, then soak plastic in ethanol for 30 mins, UV treatment and heat treatment performed). In this study, the biodegradation of polythene bag was analyzed 1 month of incubation in liquid culture method. The microbial species found associated with the degrading materials were identified as five Gram positive Bacillus, five Gram negative bacteria Bacillus, gram positive coccobacillus, one gram negative coccobacillus. The isolated organism were Bacillus subtilis, Bacillus amyloliquificiam, Bacillus amylolyticus, and Arthobacter defluvii. Then make a consortium of gram negative and gram positive organism. Gram negative organisms were Pseudomonas sp., E.coli Enterobacter. The weight loss of biodegradable plastics was 8.6%, plastic gloves use in lab weight loss percentage was 40.7% and weight loss in shopping bags used in daily life.
